MOTS-C is a 16 amino acid mitochondrial-derived peptide encoded within the 12S rRNA gene of the mitochondrial genome. Unlike nuclear-encoded peptides, MOTS-C is expressed directly from mitochondrial DNA, positioning it as a key subject in mitochondrial-nuclear communication research. Research has investigated MOTS-C's role in AMPK pathway activation, insulin sensitivity modelling, and metabolic homeostasis. In-vitro and preclinical studies have examined its capacity to regulate glucose utilisation via the folate cycle and purine biosynthesis pathways, independent of canonical insulin signalling. Further research has explored its activity as an exercise-responsive peptide, with expression observed to increase in skeletal muscle and plasma under physical stress conditions. MOTS-C has also been studied in the context of cellular ageing, with research suggesting involvement in stress resistance and mitochondrial biogenesis regulation.
