Ipamorelin is a synthetic pentapeptide growth hormone secretagogue (GHS) with the sequence Aib-His-D-2-Nal-D-Phe-Lys-NH2, identified by CAS number 170851-70-4. It acts as a selective agonist at the growth hormone secretagogue receptor 1a (GHS-R1a) — the same receptor targeted by endogenous ghrelin — and has been characterised in preclinical models for its capacity to stimulate growth hormone release from pituitary somatotrophs. Ipamorelin is distinguished within the GHRP class by its receptor selectivity: research has reported growth hormone release without the concurrent elevation of cortisol, prolactin or ACTH observed with earlier secretagogues such as GHRP-6 and hexarelin. Because it signals through the GHS-R1a and phospholipase C pathway rather than the GHRH receptor's cAMP cascade, it is frequently studied alongside GHRH analogues in synergy research, where the two independent cascades produce a greater-than-additive growth hormone response.
